Rna extraction buffer

Purpose

General purpose buffer for RNA extraction.

Can be used for

  1. Resuspending cells after pelleting before lysis
  2. Diluting an enzymatic reaction before phenol-chloroform extraction or isopropanol precipitation.

Materials and Recipe

Component Final concentration Vol
H2O – 26.19 ml
3M NaOAc pH 5.5 300 mM 3 ml
0.5 M EDTA 1 mM 60 µl
10 % SDS 0.25% v/v 750 µl
Total – 30 ml
  • Make sure that the 10% SDS stock is not precipitated. If so, warm it at 37℃ until it is dissolved before using.
