General usage of Leica microscope + troubleshooting tips

  • Turning on the scope:
  • Turn on microscope, air pump, light source, laser source (all 4 are under the desk), as well as the CO2/air-temp thing (connected altogether with one switch) on the table to the side (5 components total)
    • Wait a few seconds before turning on the next component
  • Then turn on the camera. Since the camera is the most sensitive, we turn on all other components first.
  • Turn on computer and login
  • Micro-Manager 1.4
  • Use the most recent config settings (currently 2019)
  • Power-cycle LED bars on main micro-manager dialogue box
  • Turning off the scope:
  • Turn off the 4 components under the scope and the CO2 box to the side, then turn off the camera
  • Follow instructions here for transferring images or turning on linux ssh to transfer later
  • Troubleshooting:

  • COM errors
    • These error messages typically appear when attempting to load a config file in MicroManager at startup and indicates a connection error between computer and one of the 6 components. This can typically be addressed by power-cycling all 6 components and computer then trying again.
    • It is important not to move or disconnect any USB connections on any of the devices! This will break the connection to the computer and cause a COM error. If a disrupted USB connection is the cause of the COM error, you may see additional error messages regarding a "Serial Port" or "VCP" being unavailable in the Hardware Configuration Wizard.
